# Why attach CJC-1295 to albumin?

> Albumin and duration — Idaho Hormones — At Idaho Hormones, the CJC-1295 duration essay follows albumin binding from rat experiments to the hormone response measured in healthy adults.

**Duration**

The attachment deserves the display case: a chemical design that changes the length of the experiment.

## The albumin attachment

CJC-1295 emerged from [work on growth-hormone-releasing-factor analogs that form a covalent bond with circulating serum albumin](https://pubmed.ncbi.nlm.nih.gov/15817669/). A covalent bond is a chemical attachment. Albumin supplies the carrier in this design; the peptide supplies the hormone-releasing activity. The same work tested stabilizing substitutions in the peptide.

There is a pleasing thrift to the design. Rather than treating persistence as a mystery, the experiment gives it a physical feature to investigate. The attachment and the hormone response can be examined together. It also makes the precise molecule important: the evidence follows the chemical design used in the experiment.

## The rat experiment

In rats, the [albumin-binding lead produced a 4-fold increase in growth-hormone AUC over 2 hours compared with unconjugated hGRF(1-29)](https://pubmed.ncbi.nlm.nih.gov/15817669/). AUC, the area under the concentration curve, adds up exposure across the observation period. The study also reported peptide detectable in plasma beyond 72 hours and greater stability against the enzyme dipeptidylpeptidase-IV in laboratory testing.

That is a preclinical result, with rats and laboratory assays doing the work. The comparison tests a concrete design choice, which is why I rate it. Carrying the same numerical response into a human prediction would outrun this experiment. The healthy-adult study provides its own measurements.

## The healthy-adult measurements

In healthy adults, the [estimated CJC-1295 half-life was 5.8–8.1 days](https://pubmed.ncbi.nlm.nih.gov/16352683/). Half-life describes the time for the circulating amount to fall by half. In that study, mean GH remained raised for 6 days or more and IGF-1 for 9–11 days after single administration; after multiple administrations, IGF-1 remained above baseline for up to 28 days.

The plate separates the single-administration observations from the multiple-administration observation. I would hate to lose that distinction in the appeal of the longest number. The persistence of the compound, the duration of the GH response and the duration of the IGF-1 response are separate entries in the record.

This is where duration becomes a more interesting question for me. A long-lived stimulus might sound as though it would make secretion continuous. The [pulse experiment](/pulse-record) tests what happened to the rhythm.

*Healthy-adult response windows from the [Teichman study](https://pubmed.ncbi.nlm.nih.gov/16352683/); the longer IGF-1 window followed multiple administrations.*
